Seawater Cultured
Help with making up some agar?
I am looking into making up some ZoBell 2216E agar (similar to marine agar), which contains…
0.5% polypeptone
0.1% yeast extract
1.5% agar
75% seawater
And the final agar solution will have a pH of 7.5.
This may seem like a silly question, but could anybody help me to quantitate the above formula? Say I wanted to make 1 litre of agar, how much of each compound would I need to use? I think I know what to do, but I really don’t want to mess it up – I have to culture some very special bacteria and don’t want to run the risk of killing them off!

I was wondering about the other 22.9%, but according to the journal I wish to follow (and after contacting the person who also wrote the journal AND will be sending me the bacteria that I require), he again repeated this formula for the agar. I also need to add 20 microgram per mL of chloramphenicol (in final), but this shouldn’t interfere with making up the agar itself.
Like in most growth mediums the percents represent what is not water (that is DW or DDW).
Which means you have to complete the volume (after adding all of the ingredients) to 1l.
the way to do it would be:
750 ml seawater.
15g agar
1g yeast extract
5g polypeptone.
complete to 1l with DDW.
adjust pH.
sterilize (using autoclave).
add chloramphenicol.
